摘要: A phosphate-eliminated nonnatural oligonucleotide serves as a primer surrogate in reverse transcription reaction of mRNA. Despite the triazole linkages surrogate, transcriptase effectively elongated cDNA sequences on 3'-downstream by complementary sequence structure-activity comparison with reference natural oligonucleotides shows superior priming activity containing triazole-linkages. The also protect transcribed from digestion reactions 5'-exonuclease and enable us to remove noise transcripts unknown origins.
