摘要: This chapter presents a protocol for the phylogenetic identification of microorganisms in environmental samples (water and sediments) by means fluorescence situ hybridization (FISH) with ribosomal RNA-targeted oligonucleotide probes signal amplification (catalyzed reporter deposition [CARD]). The FISH are labeled enzyme, horseradish peroxidase (HRP). A subsequent fluorescently tyramides results substantially higher intensities target cells than after directly fluorochromes. Sample preparation cell permeabilization strategies various microbial wall types discussed. custom labeling different fluorochromes is described. sequential multicolor CARD-FISH outlined simultaneous detection groups.
