Metabolic Alterations and the Protective Effect of Punicalagin Against Glutamate-Induced Oxidative Toxicity in HT22 Cells.

摘要: Oxidative stress is involved in many neurological diseases, including Alzheimer's disease. Punicalagin (PC) a hydrolysable polyphenol derived from Punica granatum and potent antioxidant. In this study, the neuroprotective effect of PC on glutamate-induced oxidative was evaluated mouse hippocampal cell line, HT22. treatment protected HT22 cells death concentration-dependent manner, potentially attenuated intracellular reactive oxygen species (ROS) restored mitochondrial membrane depolarization. Metabolic alterations after protective were with HPLC GC-MS profiling methods multivariate statistical analyses. Alterations ten metabolites identified, amino acids, aspartic acid, asparagine, threonine, anserine, cysteine, tryptophan, lysine, as well fatty acids palmitic stearic palmitoleic acid. pathway analysis revealed involvement multiple affected pathways, such cysteine methionine metabolism, tryptophan alanine, aspartate, glutamate acid oxidation. These results clearly demonstrate that promising therapeutic agent for stress-associated diseases.
