Persistent neuronal labeling by retrograde fluorescent tracers: a comparison between Fast Blue, Fluoro-Gold and various dextran conjugates

摘要: The permanence of retrograde neuronal labeling by the fluorescent tracers Fast Blue, Fluoro-Gold, Mini-Ruby, Fluoro-Ruby and Fluoro-Emerald was investigated in adult rat spinal motorneurons at 1, 4, 12 24 weeks after tracer application to a transected muscle nerve. After 1 week, largest number retrogradely labeled motoneurons found with Fluoro-Gold Fluoro-Ruby, while yielded smaller cells. With increasing survival time, all these exhibited marked decrease neurons. Blue also produced very efficient staining week and, addition, Blue-labeled cells remained constant over entire time period studied. Also embryonic cord tissue exposed Blue. label persisted for least 6 months transplantation into cord. Double-labeling experiments combining or showed that substances were non-toxic time-related neurons latter due degradation leakage dyes. Thus, would be choice motoneuronal long-term experiments, whereas usage other should restricted limited duration.
