In-Depth Investigation of Archival and Prospectively Collected Samples Reveals No Evidence for XMRV Infection in Prostate Cancer

摘要: XMRV, or xenotropic murine leukemia virus (MLV)-related virus, is a novel gammaretrovirus originally identified in studies that analyzed tissue from prostate cancer patients 2006 and blood with chronic fatigue syndrome (CFS) 2009. However, large number of subsequent failed to confirm link between XMRV infection CFS cancer. On the contrary, recent evidence indicates contaminant originating recombination two mouse endogenous retroviruses during passaging tumor xenograft (CWR22) mice, generating laboratory-derived cell lines are XMRV-infected. To refute an association cancer, we tissues plasma prospectively collected cohort 39 as well archival RNA original study. Despite comprehensive microarray, PCR, FISH, serological testing, was not detected any newly samples tissue, although remained XMRV-positive. Notably, VP62 which prototype strain derived, tested negative for on re-analysis. Analysis viral genomic human mitochondrial sequences revealed all previously characterized strains identical had been contaminated by XMRV-infected laboratory line. These findings reveal no underscore conclusion naturally acquired infection.
