摘要: Major histocompatibility complex (MHC) plays an important role in the immune response of vertebrates. In this study, we isolated MHC class IIA and IIB genes from blunt snout bream (Megalobrama amblycephala) by rapid amplification cDNA ends polymerase chain reaction (RACE-PCR). order to study function M. amblycephala, tissue distribution bacterial challenge were analyzed. All characteristic features II structure could be identified deduced amino sequences IIB, including leader peptide, α1/β1 α2/β2 domains, connecting peptide transmembrane cytoplasmic regions, as well conserved cysteines N-glycosylation site. The acid sequence molecules shared 48% 88% 65% 77% similarity with those other teleosts, respectively. Quantitative real-time PCR (qRT-PCR) demonstrated that I ubiquitously expressed ten tissues, high level related kidney, intestine, gill spleen. Challenge amblycephala extracellular pathogen, Aeromonas hydrophila, resulted a significant increase expression I, mRNA within 72 h after infection gill, intestine liver, followed recovery normal 120 h. changes levels for most tissues significantly higher than corresponding at time points (P < 0.05). These results played amblycephala; however, showed different functional activity, which need further investigated teleost.
