摘要: Epigenetic modification of the cytosine 5-position is an important regulator gene expression with essential roles in genome stability, development, and disease. In addition to 5-methylcytosine (mC), oxidized mC derivatives 5-hydroxymethyl-, 5-formyl-, 5-carboxylcytosine (hmC, fC, caC) have recently been discovered. These are intermediates active demethylation pathway but might also represent new epigenetic marks individual biological roles. This increase chemical complexity DNA-encoded information has created a pressing need for approaches that allow reading editing this information. Transcription-activator-like effectors (TALEs) DNA-binding domains programmable sequence selectivity enable direct modifications can guide enzymatic genomic loci choice. Here, we review recent advances employing TALEs these applications.
