摘要: Abstract The chromatin remodeling protein, dIno80 (Drosophila Ino80) regulates homeotic genes. We show that Ino80, along with Trx and ETP (Enhancer of Trithorax Polycomb) proteins, interacts two Polycomb/Trithorax Responsive Elements (PRE/TRE), iab-7 bxd PRE in flies the larval imaginal discs. In S2 cells, localizes to endogenous bxd-PREs. localization Ino80 Pleiohomeotic (Pho) at is sensitive cellular abundance each other; when levels are limiting, there increased Pho enrichment, knock-down leads enrichment Ino80. demonstrate over-expression rescues pupal lethality pleiohomeotic (pho) deficient flies, which suggests has a role memory. apparent competition between for binding indicates may act as potential recruiter regulatory complex addition being remodeler. Author Summary null mutants different stages development fly, implying they function independent other. observation Pho-lethality can be rescued by overexpression significant penetrance its own DNA domain, led us predict have Pho-independent functions, perhaps through non-canonical complexes. current study, we cooperation Polycomb proteins regulate effect or mutation results altered phenotype adult rescue Lac-Z expression discs, parallel similar knock-down. provide evidence direct interaction iab7- bxd-PRE using immunoprecipitation. around sequence was enhanced absence Pho, indicating PRE.
