Multi-lineage expansion potential of primitive hematopoietic progenitors: superiority of umbilical cord blood compared to mobilized peripheral blood.

摘要: Abstract Objective The majority of studies assessing ex-vivo expansion primitive hematopoietic cells only address production myeloid progeny whereas it may be more appropriate to maintain or expand progenitors that retain capacity for multilineage differentiation. In this study, we assessed the murine fetal liver cell line AFT024 (LTC-IC) and lymphoid (NK-IC) from umbilical cord blood (CB) mobilized peripheral (PB) CD34 + lin − 38 cells. Methods Sorted were established in cultures direct contact with feeder a transwell above (noncontact culture) various combinations Flt-3L (FL), stem factor, interleukin 7, thrombopoietin (Tpo), macrophage inflammatory protein-1α added. Frequency LTC-IC NK-IC was at day 0 following 2 5 weeks culture. Results CB contained significantly showed an enhanced compared PB. combination FL Tpo maximal both noncontact conditions. contrast, PB required multiple cytokines. Conclusions These results demonstrate can expanded findings emphasize importance multi-lineage differentiation expansion. Elucidation specific factors necessary will contribute development clinically applicable system.
