摘要: The transport of amino acid across the membranes has great importance in cell metabolism. Specific experimental methodologies are required for measuring vectorial reactions catalyzed by membrane transporters. So far, most widely used technique to study was measure flux intact systems expressing a specific transporter. Some limitations this procedure caused presence endogenous transporters and intracellular enzymes inaccessibility compartment. Alternative strategies which allow reducing interferences improving handling internal compartment would be useful knowledge.An protocol, makes use liposomes mediated glutamine/amino (ASCT2) transporter, solubilized from rat kidney brush borders, is described. based on reconstitution transporter removal detergent mixed micelles detergent, protein, phospholipid. assayed formed proteoliposomes Na(+) dependent uptake L: -[(3)H]glutamine antiport with -glutamine. This method allows activity under well controlled conditions permits performing experiments cannot realized systems.
