摘要: Type I collagen is the most abundant type in soft tissues and only found mineralized bone. We established a rapid equilibrium radioimmunoassay for carboxyterminal propeptide of human procollagen (PICP), to be used as an indicator synthesis collagen. isolated from medium primary cultures skin fibroblasts, digested protein with highly purified bacterial collagenase, PICP by lectin-affinity chromatography, gel filtration, ion-exchange separation on HPLC. The purity was verified sodium dodecyl sulfate-polyacrylamide electrophoresis N-terminal amino acid sequencing its component chains. final polyclonal rabbit antibodies. Material antigenically related readily detected serum. There one form serum antigen, molecular size affinity antibodies being similar those propeptide. Intra- interassay CVs are 3% 5%, respectively. Preliminary reference intervals healthy adults (18 61 years age) 38-202 micrograms/L men 50-170 women: concentration inversely age. antigen stable during storage after repeated thawing.
