摘要: Abstract A protocol is offered for the isolation of carboxyterminal propeptides human type I procollagen and development an antibody specific these propeptides. Type was harvested from media cultured fibroblasts. Digestion with bacterial collagenase released a fragment that isolated by ion-exchange chromatography. The contained telopeptides joined to could be cleaved carboxyl peptidase. Rabbit antibodies raised colt agenase-generated were sequentially adsorbed on affinity columns reference antigen collagen. obtained shown sensitive radioimmunoassays recognize conformational propeptide determinants not react triple helical telopeptide Indirect immunofluorescence indirect immunoperoxidase staining fibroblasts localized in cytoplasm, at cell surface, extracellular matrix. radioimmunoassay same has reported altered concentrations sera patients diseases affecting collagen metabolism (Taubman, et al., 1976; Savolainen 1984; Carey 1985).
