摘要: Poly(ADP-ribosyl)ation is a post-translational modification of proteins. Upon DNA damage, poly(ADP-ribose) polymerase-1 catalyzes the transfer ADP-ribose moieties from NAD+ onto acceptor proteins to form long and branched polymers. an extensive but transient as polymer chains can reach more than 200 units on protein acceptors be degraded within few minutes by glycohydrolase. Homeostasis thought play important function in cellular processes. The importance pADPR synthesis has been established vitro vivo using chemical inhibitors genetically engineered mutant mice devoid main synthesizing enzyme, PARP-1. However, PARG remains elusive. This chapter describes generation characterization knockout mice.
