摘要: We have reported earlier that immunodepletion of extrinsic pathway inhibitor (EPI) sensitizes rabbits to disseminated intravascular coagulation (DIC) induced by infusing a low concentration tissue factor (TF). now describe the effect EPI in administered endotoxin. Cortisone-treated were anti-rabbit immunoglobulin (IgG) or Fab fragments control nonimmune material before an injection In four seven anti-EPI, plasma activity levels reduced 70% 80% initial for 6 8 hours. these endotoxin extensive DIC, as evidenced substantial decreases fibrinogen, V, VIII, and platelets, gross hemorrhagic necrosis kidneys due massive deposition fibrin glomerular microcirculation (the generalized Shwartzman reaction). three anti- EPI, only transiently reduced. IgG Fab, minimal moderate clotting deposits not found capillaries. Because it is believed TF expressed on monocytes triggers endotoxin-induced coagulation, data are taken evidence functions natural anticoagulant can regulate VIIa/TF cell surfaces vivo. They support hypothesis prevents thrombotic complications might otherwise result from exposure blood cytokine-induced generation small amounts many inflammatory infectious disease states.
