摘要: The neutral N-glycan structures of Arion lusitanicus (gastropod) skin, viscera and egg glycoproteins were examined after proteolytic digestion, release the glycans from peptides, fluorescent labelling with 2-aminopyridine fractionation by charge, size hydrophobicity to obtain pure glycan structures. positions linkages sugars in analysed two dimensional HPLC (size hydrophobicity) MALDI-TOF mass spectrometry before digestion specific exoglycosidases. most striking feature adult tissues was high amount oligomannosidic small paucimannosidic terminated 3-O-methylated mannoses. truncated often contained modifications inner core β1,2-linked xylose β-mannose residue and/or an α-fucosylation (mainly α1,6-) innermost GlcNAc residue. Skin showed predominantly same glycans, however, different amounts. Traces large carrying galactoses also detected. mainly (≈ 75%) some modified or α1,6-fucose, but this case no methylation any monosaccharide Thus, gastropods seem be capable producing many types ranging those typical human similar found nematodes, therefore will a valuable model understand regulation glycosylation. Furthermore, opens way for using organism as host production recombinant proteins. detailed knowledge on glycosylation may help identify targets pest control.
