摘要: High-frequency, synchronous embryogenic systems in liquid culture facilitate plant regeneration and can be used as an essential model for performing functional genomics studies understanding molecular aspect of the ontogenesis higher plants. In present study, somatic cultures were developed Muscadinia rotundifolia cv. Darlene Vitis vinifera Velika. High cell density presence 2,4-dichlorophenoxyacetic acid (2,4-D) proved to establishment suspension cultures. Low continuous availability auxin was crucial maintenance withdrawal 2,4-D is sufficient advance embryo development toward differentiation plantlets regeneration. Cells clusters fractionation by gradient centrifugation Ficoll solution demonstrated a suitable method separation subpopulations with various potential development. The high frequency embryos attained essentially heaviest (at 16-18 % >18 layer) population.
