The DSCs-Expressed CD82 Controls the Invasiveness of Trophoblast Cells via Integrinbeta1/MAPK/MAPK3/1 Signaling Pathway in Human First-Trimester Pregnancy

摘要: CD82 is recognized as a wide-spectrum tumor metastasis suppressor that inhibits cancer cell motility and invasiveness. At the human maternal-fetal interface, decidua believed to effectively limit inappropriate invasion of trophoblasts. Here we have found transcription translation in decidual stromal cells (DSCs), whereas trophoblast do not express CD82. The in-cell Western analysis reveals attenuation DSCs by chorionic gonadotropin (hCG), but estrogen or progesterone. It demonstrated silencing RNA interference increases integrinb1, decreases TIMP1 expression DSCs, promotes first-trimester trophoblasts coculture. Moreover, U0126, anti-integrinb1 neutralizing antibody, reverses decreased increased invasiveness cells, antibody also MAPK3/1 phosphorylation induced silence. After transfection with CD82, invasive index BeWo significantly increase. results above indicate DSCs-expressed up-regulates an autocrine manner partly through integrinb1/MAPK/MAPK3/1 signaling pathway. Furthermore, mRNA protein level miscarriage higher than normal early pregnancy, which implies abnormal restricts appropriate leads pregnancy wastage.
