Cognitive performances of cholinergically depleted rats following chronic donepezil administration

摘要: Since acute and chronic administration of the acetylcholinesterase inhibitors, namely donepezil, improves cognitive functions in patients afflicted by mild to moderate dementia reverses memory deficits experimental models learning memory, it seemed interesting assess effects donepezil treatment on adult rats with forebrain cholinergic depletion. Lesions were performed means intracerebroventricular injections immunotoxin 192 IgG-saporin. The lesioned animals treated or not compared those intact animals. Cholinergic depletion affected working functions, weakened procedural competencies, acquisition localizing knowledge, evoked remarkable compulsive perseverative behaviors. In animals, ameliorated localizatory capabilities, performances linked flexibility abilities. Furthermore, attenuated deficits. present data indicate positive specific performances, suggesting that an aimed use targeting some symptoms more than others, may be beneficial case hypofunction. animal model used research provide efficient method for analyzing cognition-enhancing drugs before clinical trials.
