Telomerase inhibition and telomere loss in BEL-7404 human hepatoma cells treated with doxorubicin

摘要: AIM: To study the effects of doxorubicin on telomerase activity and telomere length in hepatocellular carcinoma. METHODS: Telomerase was assayed with a non-radioisotopic quantitative repeat amplification protocal-based method. The effect (DOX) growth BEL-7404 human hepatoma cells determined by microculture tetrazolium assay. Mean (terminal restriction fragment) detected Southern blot expression subunits genes investigated RT-PCR. Cell apoptosis cell cycle distribution were evaluated flow cytometry. RESULTS: inhibited dose time-dependent manner treated DOX for 24, 48 or 72 h concentrations from 0.156 to 2.5 μM which crrelated inhibition growth. No changes found mRNA three (hTERT, hTR TP1) after drug exposure indicated concentrations. showed shortened mean accumulated at G2/M phase. However, there almost no DOX. CONCLUSION: shortening may contribute its efficiency treatment carcinoma.
