摘要: Three-dimensional (3D) in vitro models have an invaluable role understanding the behaviour of tumour cells a well defined microenvironment. This is because some aspects characteristics cannot be fully recapitulated cell monolayer (2D). In present study, we compared growth patterns, expression signalling molecules, and metabolism-associated proteins endometrial cancer lines 3D 2D cultures. Cancer formed spherical structures reconstituted basement membrane (3D rBM), morphological appearance was line dependent. Cell differentiation observed after 8 days rBM. There reduced proliferation, detected by less PCNA rBM than monolayers. The addition exogenous epidermal factor (EGF) to induced phosphorylation EGFR Akt both culture conditions. uptake glucose selectively altered rBM, but there lack association with Glut-1 expression. secretion vascular endothelial (VEGF) prostaglandin E(2) (PGE(2)) it Our data demonstrated that as model can influence proliferation metabolism monolayer. Changes are specific individual types. use is, therefore, important study targeted anticancer therapies.
