摘要: We characterized a novel guanine nucleotide exchange factor (GEF) for Ras family G proteins that is highly homologous to CalDAG-GEFI, GEF Rap1 and R-Ras, RasGRP/CalDAG-GEFII, R-Ras. This GEF, referred as CalDAG-GEFIII, increased the GTP/GDP ratio of Ha-Ras, in 293T cells. CalDAG-GEFIII promoted vitroalso, indicating exhibited widest substrate specificity among known GEFs proteins. Expression was detected glial cells brain glomerular mesangial kidney by situhybridization. activated ERK/MAPK most efficiently, followed CalDAG-GEFII CalDAG-GEFI JNK activation prominent expressing CalDAG-GEFII, CalDAG-GEFI. induced neuronal differentiation PC12 anchorage-independent growth Rat1A less efficiently than did CalDAG-GEFII. Thus, co-activation apparently attenuated Ras-MAPK-dependent cellular transformation. Altogether, broad range biological activity different from either or
