摘要: Background and Design: Palmoplantar pustulosis (PPP) is an inflammatory skin disease characterized by pustule formation, erythema, induration, scaling of the affected palms soles. usually resistant to treatment. In a double-blind study (phase 1) 4 weeks, 40 patients with PPP were randomized receive oral cyclosporine, 2.5 mg/kg per day, or placebo. An open-label dose-finding phase 2 cyclosporine doses 1.25, 2.5, 3.75 day was performed in following 3 months. The then followed for at least months after termination Response treatment judged number fresh pustules. Patients displaying reduction 50% greater pustules, compared baseline, defined as responders. Results: Of who completed 1, 17 19 group four 15 placebo classified responders ( P =.001). subsequent open phase, daily dose 1.25 appeared be effective approximately half treated patients. Many relapsed initial success cyclosporine. However, only one patient studied totally failed respond At end 3, most parameters had returned pretreatment levels. common side effect headache dosage group; no significant effects observed group. Conclusions: Low-dose (1.25 day) PPP. Arch Dermatol. 1993;129:1273-1279)
