摘要: Before cells can invade the extracellular matrix, they must alter their expression of adhesion receptors. Syndecan-1 is a cell surface proteoglycan that binds to matrix and undergoes changes in during development, differentiation some types, following malignant transformation tumors. To determine if syndecan-1 influence invasion, we employed model system which human B lymphoid type I collagen gels. Examination panel lines reveals those not expressing migrate within collagen. In contrast, fail invade. directly assess effect on ARH-77 cells, do express readily collagen, were transfected with cDNA for syndecan-1. The syndecan-1-positive transfectants exhibit drastically reduced ability as compared parental cells. This inhibition invasion by reversed preincubating gels heparin or growing chlorate, an inhibitor glycosaminoglycan sulfation. These results demonstrate inhibits into loss may be necessary prior migration normal metastatic
