Effects of cytosine arabinoside and hydroxyurea on the synthesis of deoxyribonucleotides and DNA replication in L1210 cells.

摘要: Experiments were carried out in L1210 cells to examine the importance of 'substrate cycles' regulating intracellular levels deoxyribonucleoside 5'-triphosphate. incubated with [14C]cytidine or [14C]adenosine presence and absence hydroxyurea cytosine arabinoside (araC). These incubations for either 30 120 min. Inhibition ribonucleotide reductase by resulted blockage flux ribonucleotides deoxyribonucleotides (greater than 90%) as expected. When DNA synthesis was inhibited araC, there a marked decrease incorporation into deoxyribonucleotides. However, not corresponding increase deoxyribonucleotide acid-soluble fraction deoxyribonucleosides culture medium. AraC treatment decreased total formation data indicate that do regulate pools dNTPs via which involve activation phosphatases when is blocked kinases inhibited.
