Post-translational Mechanisms Regulating NK Cell Activating Receptors and Their Ligands in Cancer: Potential Targets for Therapeutic Intervention.

摘要: Efficient clearance of transformed cells by Natural Killer (NK) is regulated several activating receptors, including NKG2D, NCRs, and DNAM-1. Expression these receptors as well their specific "induced self" ligands finely during malignant transformation through the integration different mechanisms acting on transcriptional, post-transcriptional, post-translational levels. Among mechanisms, release in extracellular milieu protease-mediated cleavage or vesicle secretion represents some relevant cancer immune escape processes. Moreover, covalent modifications ubiquitination SUMOylation also contribute to negative regulation NKG2D DNAM-1 ligand surface expression resulting either intracellular retention and/or degradation. All greatly impact NK cell mediated recognition killing may be targeted potentiate surveillance against tumors. Our mini review summarizes main regulating with particular emphasis contribution shedding ubiquitin ubiquitin-like reducing target susceptibility cell-mediated killing. Strategies aimed at inhibiting order preserve will discussed.
