Novel amplicons on the short arm of chromosome 7 identified using high resolution array CGH contain over expressed genes in addition to EGFR in glioblastoma multiforme

摘要: Amplification of a defined chromosome segment on the short arm 7 has frequently been reported in glioblastoma multiforme (GBM), where it is generally assumed that result over expression epidermal growth factor receptor (EGFR) gene provides selective pressure to maintain amplification event. We have used high resolution array comparative genomic hybridization (aCGH) analyze events 7p GBM, which demonstrates that, fact, several other regions distinct from EGFR can be amplified. To determine changes levels associated with these events, we oligonucleotide arrays investigate genes amplified were also expressed. These analyses demonstrated not all amplicons showed increased expression, and series expressed could potentially contribute development malignant phenotype tumors. The global analysis afforded by aCGH improved our ability define numerical abnormalities cancer cells raised possibility than may important.
