摘要: Abstract A simple, sensitive and reliable ultra fast liquid chromatography-electrospray ionization–tandem mass spectrometry (UFLC-ESI–MS/MS) method was developed for simultaneously quantifying gastrodin ( p -hydroxy-methyl-phenol-β- d -glucoside) its metabolite -hydroxybenzyl alcohol (HBA) in dog plasma. Separation performed on an chromatography (UFLC) system. Detection carried out a tandem (MS/MS) multiple reaction monitoring (MRM) mode via electrospray ionization (ESI) interface. MRM of precursor–product ion transitions used gastrodin, HBA the internal standard (IS, bergeninum) at m/z 285.0 → 123.0, 123.0 → 105.0 326.9 → 192.2, respectively. The lower limits quantification (LLOQ) this both were 1 ng/mL, with their linear concentration ranging from 0.001 to 10 μg/mL. methods validated selectivity, calibration curves, accuracy precision, extraction recoveries, matrix effects, carry-over, cross talk, dilution integrity, stability incurred sample reanalysis (ISR). Using method, pharmacokinetic behaviors after intragastric administration (ig) dogs studied first time.
