摘要: Abstract A chimeric porcine circovirus type 1-2 (PCV1-2) infectious DNA clone has low transfection efficiency and exhibits levels of proliferation. Electroporation lipofection parameters were optimized for PK-15 Dulac cells with the purpose increasing rescuing PCV1-2. Titers PCV1-2 in 100-fold higher than those following transfection. The electroporation into was high when three 400 μs pulses at 250 V 6 μg plasmid used, ratio to reagent 1:3. proportion infected 55.6% compared 44.2%, techniques respectively. Virus titers cells, from 104.44 105.32 TCID50/mL 101.90–103.38 TCID50/mL cells. more permissive regardless technique.
