SIMULTANEOUS DETERMINATION OF COCAINE, COCAETHYLENE, AND THEIR POSSIBLE PENTAFLUOROPROPYLATED METABOLITES AND PYROLYSIS PRODUCTS BY GAS CHROMATOGRAPHY/MASS SPECTROMETRY

摘要: During the investigations of fatal transportation accidents, samples from victims are also analyzed for drugs, including cocaine. Cocaine is abused by smoking, nasal insufflantion, and intravenous injection, it taken with ethanol. Therefore, important to determine concentrations cocaine its metabolites, ethanol analogs, pyrolysis products establishing degree toxicity, possible ingestion ethanol, route administration. In this study, a sensitive selective procedure developed simultaneous analyses cocaine, benzoylecgonine, norbenzoylecgonine, norcocaine, ecgonine, ecgonine methyl ester, m-hydroxybenzoylecgonine, anhydroecgonine ester (AEME), (AECG), cocaethylene, norcocaethylene, ethyl in blood, urine, muscle homogenate. analysis, available deuterated analogs these analytes were used as internal standards. Proteins blood homogenate precipitated cold acetonitrile. After removal acetonitrile evaporation, supernatants urine extracted solid-phase chromatography. The eluted converted hydrochloride salts derivatized pentafluoropropionic anhydride 2,2,3,3,3-pentafluoro-1-propanol. on gas chromatograph (GC)/mass spectrometer system selected ion monitoring. This method was successfully applied analyzing 13 case specimens aviation accident pilot fatalities and/or motor vehicle operators. AEME found consistent those produced solely GC inlet controls, suggesting that not cases smoking. Although remains potential marker abuse AECG useful because low recovery production metabolites. unique multiple can be solid tissues single extraction increased sensitivity through formation using one-step derivatization.
