摘要: Purpose:To determine whether altering the dietary content of N-6 (n-6) and N-3 (n-3) polyunsat- urated fatty acids affects growth androgen-sensitive prostate cancer xenografts, tumor membrane acid composition, cyclooxygenase-2 prostaglandin E2 (PGE2) levels. Experimental Design: Individually caged male severe combined immunodeficiency mice were fed isocaloric 20% kcal fat diets with derived either primarily from n-6 (n-6 group) or consisting n-3 in a ratio 1:1 (n-3 group), injected s.c. Los Angeles Prostate Cancer 4 (LAPC-4) cells. Tumor volumes mouse weights measured weekly, caloric intake was 3 days per week, tumors serum harvested at 8 weeks postinjection. Results: rates, finaltumor vol umes, prostate-specific antigen l evel s reduced group relative to group. The had decreased proliferation (Ki67 staining) increased apoptosis (terminal nucleotidyl transferase ^mediated nick end labeling staining). In vitro LAPC-4 cells medium containing by 22% cultured group. The n-6/n-3 ratios membranes lower addition, protein mRNA levels, an 83% reduction PGE2 vascular endothelial factor expression. Conclusion: These results provide sound basis for clinical trials evaluating effect fish oilon PGE 2 biomarkers progression men cancer.
