摘要: Replication-independent histone variants can replace the canonical replication-dependent histones. Vertebrates have multiple H2A variant histones, including H2AZ and H2AX that are present in most eukaryotes. regulates transcriptional activation as well maintenance of gene silencing, while is important DNA damage repair. The fruit fly Drosophila melanogaster has only one (H2AV), which a chimera H2AX. In this study we found lack H2AV led to formation black melanotic masses third instar larvae. these was conjunction with loss majority primary lymph gland lobes. Interestingly, cells posterior signaling center were preserved mutants. Reduction levels by RNAi knockdown caused milder phenotype structure but included precocious differentiation prohemocytes located within medullary zone secondary lobes gland. Mutant rescue experiments suggest H2AZ-like rather than H2AX-like function primarily required for normal hematopoiesis.
