摘要: This research aimed to develop a supercritical fluid (SCF) technique for preparing particulate form of itraconazole (ITZ) with good dissolution and bioavailability characteristics. The ITZ solid dispersion was formulated hydroxypropyl methylcellulose, Pluronic F-127, L-ascorbic acid. Aggregated particles showed porous structure when examined by scanning electron microscopy. Powder X-ray diffraction Fourier transform infrared spectra indicated an interaction between excipients that existed in amorphous state the composite particles. obtained SCF process improved media pH 1.0, 4.5, 6.8, compared commercial product (Sporanox(®)), which could be ascribed aggregated particle shape ITZ. While did not show statistical improvement (P=0.50) terms oral Sporanox(®), C max (the maximum plasma concentration pharmacokinetic curve) raised significantly (P=0.03) after administration.
