Effects of a Novel pH-sensitive Liposome With Cleavable Esterase-Catalyzed and pH-responsive Double Smart mPEG Lipid Derivative on ABC Phenomenon

摘要: Background The ABC phenomenon is described as a syndrome of accelerated clearance polyethylene glycol (PEG)-modified liposomes from the bloodstream when repeatedly injected, with their increased accumulation in liver and spleen. Methods To clarify this immune response phenomenon, we evaluated novel modified pH-sensitive liposome cleavable double smart PEG-lipid derivative (mPEG-Hz-CHEMS). Results mice was brought about by repeated injection conventional PEG-PE accompanied greatly uptake liver. However, slight mPEG-CHEMS only slightly liver, mPEG-Hz-CHEMS did not induce there no increase accumulation. This finding indicates that could lessen or eliminate induced PEGylated liposomes. Conclusion research has shed some light on solution to using PEG-Hz-CHEMS encapsulated nanoparticles.
