摘要: Background Lung transplantation is severely limited by an inadequate supply of lungs from brain-dead donors. A potential solution use non-heart-beating donors (NHBDs) with retrieval at intervals after circulatory arrest and death. warm ischemic period concomitant reperfusion injury a major limiting factor in the retrieved NHBDs. We hypothesized that administration nitric oxide-donor nitroglycerin to NHBDs would reduce ischemia-reperfusion activation guanylate cyclase form guanosine 3′,5′-cyclic monophosphate (cGMP). Methods An situ isolated perfused rat lung model was used. Lungs were rats varying either ventilated O2 or not ventilated. reperfused death Earle’s without (0.1 mg/ml). assessed capillary filtration coefficient, wet-to-dry weight ratio, pulmonary hemodynamics. Tissue levels adenine nucleotides cGMP concentrations measured high-performance liquid chromatography enzyme immunoassay, respectively. Results Reperfusion decreased coefficient compared all post-mortem times, irrespective pre-harvest ventilation. increased significantly nitroglycerin-reperfusion attenuated decreases high-energy nucleotides. Conclusions may facilitate safe reducing leak reperfusion.
