摘要: Abstract Background: Lungs retrieved from cadavers after death and circulatory arrest may alleviate the critical shortage of lungs for transplant. We report a rat lung transplantation model that allows serial measurement arterial blood gases left single non–heart beating donors. Methods: Twelve Sprague-Dawley rats underwent with vascular cuff technique. Donor were anesthetized intraperitoneal injection pentobarbital, heparinized, intubated via tracheotomy, then killed pentobarbital. immediately or 2 hours oxygen ventilation (tidal volume 1 mL/100 g, rate 40/min FIO = 1.0, positive end-expiratory pressure 5 cm H O). Recipient anesthetized, intubated, ventilated. The carotid artery jugular vein cannulated infusion Ringer's lactate (4 mL/h). Anesthesia was maintained halothane 0.2%, recipient measured at 4 6 snaring right pulmonary minutes. Animals put to transplantation, portions transplanted frozen in liquid nitrogen assayed wet/dry ratio, myeloperoxidase as measure neutrophil infiltration, conjugated dienes free radical–mediated lipid peroxidation. Results: Arterial PO ratio not significantly different recipients donor ventilation. Significant infiltration observed oxygenventilated Conclusions: Strategies ameliorate reperfusion injury allow successful J Thorac Cardiovasc Surg 2001;122:18-23
