摘要: BACKGROUND & OBJECTIVES There is a need to delimit the areas of filariasis transmission in view Filariasis Elimination Programme launched India. Infection rate vectors an important parameter determining and it conventionally assessed by dissection microscopy. A PCR assay based on Ssp I repeats Wuchereria bancrofti has shown potential detection infection vectors. The aim present study was evaluate specificity sensitivity this W. its vector, Culex quinquefasciatus, prevalent METHODS DNA from pools C. quinquefasciatus which microfilariae (mf) were added, extracted lysing with 0.1 M NaOH 0.2 per cent sodium dodecyl sulphate (SDS), followed silica absorption presence guanidinium thiocyanate. samples carried out using NV-1 NV-2 primers species specific SspI band visualized agarose gels stained ethidium bromide. RESULTS found be highly specific, as did not detect closely related filarial parasite, Brugia malayi. detected little 0.04 pg bancrarofti DNA. Minimum number parasite detectable mosquitoes 1 mf. pool size 50 optimum for assay. INTERPRETATION CONCLUSION sensitive detecting therefore application rapid assessment filariasis.
