Flavones from Scutellaria baicalensis georgi attenuate apoptosis and protein oxidation in neuronal cell lines

摘要: The oxidative modification of proteins plays a major role in number human diseases including Alzheimer's disease (AD). Flavones extracts Scutellaria baicalensis (SbE) have been reported to exceptional antioxidant properties. We examined the effects SbE on neuronal cells exposed stress. Neuronal HT-22 were low levels H(2)O(2) generated from glucose oxidase (GO) under conditions that caused cell death 24 h. mechanism was shown occur via apoptosis. Flavone (50 microg/ml) protected and increased viability 85+/-5% (P<0.001). flavones also content Bcl-2 cell, resulted its phosphorylation, contrast decreased Bax levels. Furthermore, oxidative-stress-induced protein carbonyl formation reduced nearly two-fold when pretreated with flavone extract. Two-dimensional electrophoresis (2-DE) showed less than 15% total visible oxidized oxidation specific for certain oxidation-sensitive proteins. These data support idea can attenuate oxidant stress protect lethal damage.
