CCAAT enhancer binding protein β has a crucial role in regulating breast cancer cell growth via activating the TGF‑β‑Smad3 signaling pathway

摘要: The aim of the present study was to examine effect CCAAT enhancer binding protein β (C/EBPβ) on human breast cancer cells. plasmids pCDH-C/EBPβ and pLKO.1-shC/EBPβ were constructed infected into MDA-MB-468 cells, provide C/EBPβ overexpressing knockdown respectively. Cell viability, cell cycle apoptosis observed by MTT assay flow cytometry analysis. Protein expression levels C/EBPβ, TGF-β1, P-Smad3 Smad3 detected western blotting. showed that absorbance cells in group increased, whereas decreased, compared with respective control at 48 72 h. Flow cytometric analysis indicated percentage G2 phase significantly increased (P<0.05) decreased group. proportion apoptotic controls. scratch-wound revealed depleted exhibited reduced motility Moreover, blotting demonstrated transforming growth factor (TGF)β1 expression, TGFβ1 levels. has a crucial role regulating through activating TGF-β-Smad3 signaling. These findings suggest may be potential therapeutic target for cancer; however, vivo studies are required confirm this.
