摘要: Peptides purified by HPLC are often in the form of a trifluoroacetate (TFA) salt, because trifluoroacetic acid is used as solvent reversed-phase separation. However, potential effects this contaminant culture systems have not been addressed previously. TFA (10(-8) to 10(-7) M) reduced cell numbers and thymidine incorporation into fetal rat osteoblast cultures after 24 h. Similar were found articular chondrocytes neonatal mouse calvariae, indicating that effect specific one type or species origin. When activities hydrochloride salts amylin, amylin-(1-8), calcitonin compared osteoblasts, proliferation was consistently less with these peptides, resulting failure detect proliferative wrongly attributing an antiproliferative effect. This finding likely be relevant all studies peptides concentrations above 10(-9) M whatever tissue type.
