摘要: Glucose transporter (GLUT) 1 is found highly expressed in malignant tumors and considered a mediator inducing cancer metastasis. Curcumin natural product which exerts anti-invasion metastasis effects cancer. This study aimed at evaluating whether attenuating GLUT1 was involved curcumin’s effects. In the vitro part, constricted pcDNA3.1-GLUT1 vector transfected into A549 cells. MTT assay used to assess on proliferation lung Transwell evaluate effect of curcumin Real-time PCR Western-blotting were employed examine expression levels GLUT1, membrane type 1-MMP (MT1-MMP) matrix metalloproteinase (MMP) 2 curcumin- incubated vivo tumor weight metastatic rate assessed nude mice bearing untransfected, empty cells originated tumors. this study, we that began show significant cytotoxicity against 45 μmol/L. inhibited invasion expressions MT1-MMP MMP2 untransfected concentration-dependent manner. exhibited resistance by up-regulating GLUT2, MMP2. Furthermore, failed decrease These results suggested inhibit GLUT1/MT1-MMP/MMP2 pathway.
