摘要: End-binding 1 protein (EB1) is a key player in the regulation of microtubule (MT) dynamics. Here, we investigated role EB1 glioblastoma (GBM) tumor progression and its potential predictive for response to Vinca-alkaloid chemotherapy. Immunohistological analysis 109 human GBM cases revealed that overexpression correlated with poor outcome including progression-free survival overall survival. Downregulation by shRNA inhibited cell migration proliferation vitro. Conversely, promoted them accelerated growth orthotopically-transplanted nude mice. Furthermore, was largely overexpressed stem-like GBM6 display vivo higher tumorigenicity more infiltrative pattern than GBM9. showed strong EB1-dependent migratory potential. The cells chemotherapy investigated. Vinflunine vincristine increased EB1-overexpressing U87 bearing mice were effective inhibit clones controls. Vinca increase MT rate length induced overexpression. Altogether, our results show expression level has prognostic value GBM, could improve treatment patients tumor.
