摘要: Using an FLP/FRT‑based method to create germline clones, we screened Drosophila chromosome arms 2L and 3R for new female meiotic mutants. The screen was designed to recover mutants with severe effects on exchange and/or segregation. This yielded 11 mutants, including six alleles of previously known genes (c(2)M ald/mps1). remaining five appear define at least four genes whose ablation results in defects. Three the novel mutants were identified molecular level. Two these, mcm5A7 tremF9, roles recombination, while a third, conaA12, is important synaptonemal complex assembly. Surprisingly, nine which lesion has been level are not result mutations characteristic EMS mutagenesis, but rather due insertion transposable element Doc. study demonstrates the utility clone‑based screens discovery strong essential genes, use genetic techniques map loci.
