2H and 13C NMR Spectroscopy of Lipid Model Membranes

摘要: The elucidation of structure and dynamics biological model membranes has been an area intensive research over the last years. As are very complex also difficult to isolate in sufficient quantities needed for certain types physicochemical techniques, many experiments have performed with defined chemical composition. While calorimetry provides information on thermodynamic properties system X-ray techniques can elucidate structural aspects bilayers, lipid molecules be elucidated by spectroscopic techniques. In particular, NMR spectroscopy is a powerful technique because it intrinsic method does not require introduction potentially perturbing label or probe as case ESR fluorescence spectroscopy. Particularly 2H l3C well suited several reasons: 1. The 13C nuclei introduced into under study via synthesis, so that either only one resonance observed (in deuterium) spectrum dominated line nucleus enriched site. 2. The shift anisotropy possibility studying motions 500-5000-Hz range. 3. The quadrupole interaction range 2 x 105 Hz deuterium spectra therefore sensitive 103-106-HZ range. 4. Simulations echo provide dynamics, but molecular order system. A combination these two give fairly complete picture motional, conformational, lipids membranes.
