摘要: The G protein-coupled receptor (GPCR) Smoothened (Smo) is the signal transducer of developmentally and therapeutically relevant Hedgehog (Hh) pathway. Although recent structural analyses have advanced our understanding Smo biology, several questions remain. Chief among them are identity its natural ligand, regulatory processes controlling activation, mechanisms by which it signals to downstream effectors. In this review, we discuss discoveries from multiple model systems that set stage for solving these mysteries. We focus on roles distinct functional domains, post-translational modifications, trafficking, conclude discussing their contributions output.
