摘要: Six healthy adult mares were each given a single IV injection of trimethoprim (TMP)-sulfamethoxazole (SMZ) at dosage 2.5 mg TMP/kg body weight and 12.5 SMZ/kg. Serum concentrations drug measured serially over 24-hour period. For TMP, the mean overall elimination rate constant (K) was 0.43/hr half-life (t1/2) 1.9 hours. The apparent volume distribution (at steady state) 1.62 L/kg TMP clearance 886 ml/hr/kg. SMZ, K 0.22/hr t1/2 3.53 state 0.33 SMZ 78.2 Each mare then 5 consecutive oral doses TMP-SMZ SMZ/kg 12-hour intervals. Trimethoprim in serum, synovial fluid, peritoneal CSF, urine, endometrium. Although both serum higher after 5th dose than 1st dose, only concentration significantly (P less 0.05) different. After attained fluids (except CSF) endometrial tissue equal to or exceeded reported minimum inhibitory for Corynebacterium pseudotuberculosis, Staphylococcus sp, Streptococcus zooepidemicus, several obligate anaerobes. Absorption drugs variable administration.
