摘要: The synthetic prostaglandin ONO-802 was administered intravaginally to Sprague Dawley rats at doses of 1.0, 0.5, and 0.125 mg/kg on days 6 through 15 gestation. A vehicle control group treated with pessaries that did not contain the drug while another remained untreated. Body weight, food, water consumption, clinical signs were monitored during experiment. In Phase One, 20 pregnant animals from each sacrificed term, major organs weighed, litter fetal data collected. Two ten dams per allowed deliver their litters, offspring evaluated for survival, growth, developmental signs, physiological function. Selected F1 retained assess learning emotional behavior or reproductive capacity. Administration either 0.5 1.0 resulted in a slight reduction food consumption body weight gain. Water increased both after dosing period mid high dose dams. Significantly weights heart, lungs, liver, adrenals, ovaries decreased thymus gland noted term sacrifice dams, whereas had adrenals only. Litter parameters unaffected by treatment. Weights female fetuses groups significantly reduced when compared controls. There no significant drug-related abnormalities among evidence treatment F0 affected development, behavior, performance offspring. Thus, teratogenic given intravaginal route.
