摘要: This study was designed to identify a suitable freezing protocol for rabbit semen by comparing the effects of different concentrations and equilibration times dimethylacetamide (DMA) dimethylsulfoxide (DMSO) on postthaw quality semen. After establishing best protocols each cryoprotectant, their efficacy compared examining in vivo fertilizing capacity samples. Pooled samples diluted medium containing 4%, 6%, or 8% DMA DMSO (all combined with 1% sucrose as nonpermeating cryoprotectant) were loaded straws equilibrated 5, 15, 45 min before liquid nitrogen vapor. The variables assessed after thawing sperm motility, viability, osmotic resistance, acrosome DNA integrity. Marked these shown cryoprotectant concentration time, results obtained using 6% min. These selected compare two cryoprotectants an insemination trial. Three groups 114 does (28 nulliparous 86 multiparous group) inseminated fresh frozen optimized protocols. Fertility rates numbers kids born similar, respectively DMSO-frozen (79.8% 7.7 ± 0.3 young per kindling) (81.6% 8.6 0.3) yet higher (P ≤ 0.05) than returned DMA-frozen (47.4% 6.7 0.4). Moreover, rabbits alive when used protocol, despite being lower those recorded semen, < 0.05). physiological status (nulliparous multiparous) had no influence fertility prolificacy results. Our findings indicate that cryosurvival is highly influenced time exposed agent freezing. According our data, emerged more effective cryopreservation sperm.
