摘要: To explore the role of calcium in mediating action carbachol chloride-secreting epithelia, we simultaneously measured intracellular free (Ca) ((Ca).) and potassium conductance (gx) basolateral membrane T84 cells grown on collagen-coated filters. (Call was with fura-2 fluores- cence microscopy expressed as a relative value ((Ca)() normalized to control. assess changes gK, short circuit current (I~ presence luminal amphotericin transepithelial mucosa-to-serosa K + gradient (Germann, W.J., M. E. Lowy, S. A. Ernst, D. C. Dawson. 1986.J. Gen. Physiol. 88:237-251). Treatment monolayers resulted parallel increase then decrease (Ca)~ gx. The carbachol-induced gg appeared be dependent (Ca)i because stimulation ofg x significantly diminished when hormone-induced blunted, either by loading BAPTA or reducing extracellular (Ca). carbachol-stimulated gK direct result steady-state (Ca)~. (C,a)~ after were correlated ionomycin also increased manner. Agi~ per A(Ca)~, however, greater than that ionomycin. Because appear open same channel, conclusion based inhibitor selectivity experiments, may have second amplifies effect
