T-lymphocyte rosette inhibition titering of antisera by direct microassay☆

摘要: A 60-sample micro-rosette inhibition assay for determining the 100% rosette inhibitory titers of heterologous antisera is described. The performed in histocompatibility trays, under oil, using frozen-thawed thymocytes or peripheral blood lymphocytes as forming lymphoid cells. Antisera dilutions are incubated with an appropriate period, either without added complement; SRBC inoculated into each well, and rosettes formed by centrifugation trays at 200 × g. Following centrifugation, glutaraldehyde to well fix bottom plates inverted allow unbound fall away. One hundred percent determined low power microscopic examination wells. Highly reproducible in-assay (±4% average standard error) between-assay (±9%) values obtained which correlated (r = 0.99) calculated more conventional methodology.
