摘要: Polyamidoamine dendrimer (PAMAM) is a new nanometer material, which can transfer the target genes to cells with high efficiency and lower toxicity. This study aims evaluate antitumor effects of survivin antisense oligonucleotide (survivin-asODN) (carried by polyamidoamine liposome) on hepatic cancer in nude mice. Hepatic model was established injecting SMMC-7721 subcutaneously into flanks liposome were mixed survivin-asODN, respectively. The shape size complex observed transmission electron microscope, zeta potential measured an analytical tool. Encapsulation DNA loading level determined ultraviolet spectrophotometer centrifuging method. Expression transplant tumor Western blotting. No significant difference appeared for diameter envelopment ratio between PAMAM liposome-survivin-asODN PAMAM-survivin-asODN (P > 0.05). Both transfection higher than that (P < 0.05). protein weight tumors transplanted group less Cell apoptosis rate In conclusion, deliver survivin-asODN effectively. Ployamidoamine liposome-mediated inhibit cell proliferation inducing apoptosis.
